Biodistribution Analysis Service

After RDC (Radionuclide Drug Conjugate) synthesis, and in vitro assays for quality control, experiments on organ level and animal models are essential. Biodistribution is a crucial point in dosimetry calculations since almost all personalized internal dosimetry is derived by projecting biodistribution measurements into dosimetry calculations.

As a professional and comprehensive large CRO company with excellent RDC analysis platform, Rdcthera is capable to conduct RDC biodistribution analysis for further clinical research.

The Importance of Biodistribution Analysis

The RDC biodistribution analysis is an essential test of the quality control to identify defects in the quality of RDC, which is usually the first indication when something might be wrong, found during an imaging procedure, especially because RDC is a novel drug for the treatment of cancer. Biodistribution study in animal models must be performed during the development of a novel radiopharmaceutical by research institutions, as an essential part of the quality assurance procedure, before clinical research on humans and laterally released for sale.

How to Perform Biodistribution Analysis

The biodistribution analysis is conducted in animals with normal phenotypes but more are being performed in animals with specific tumors by imaging the animals on either PET (Positron Emission Computed Tomography) or SPECT (Single-Photon Emission Computed Tomography) after RDC injection to measure the distribution of the radioactivity in tissues by counting or autoradiographic techniques.

For example, somatostatin receptors (SSTRs), which are overexpressed on the neuroendocrine neoplasms (NENs), can be identified by somatostatin analogs (SSAs). To evaluate the new pan-somatostatin receptors (pan-SSTRs) probe PA1, the biodistribution analysis is conducted to compare the performance between novel [64Cu]NOTA-PA1 and already discovered [64Cu]DOTA-TATE. The micro-PET imaging results of [64Cu]NOTA-PA1 and [64Cu]DOTA-TATE in A549 tumor-bearing mice show that both the tumor concentration and the tumor uptake of [64Cu]NOTA-PA1 are much higher than [64Cu]DOTA-TATE, suggesting that [64Cu]NOTA-PA1 has better performance than [64Cu]DOTA-TATE in tumor mouse models.

Our Service

  • Mouse Models for Biodistribution Analysis
  • Mouse models of cancer enable researchers to learn about tumor biology and to examine physiological interactions among and between tumor cells and their environment in a complicated and dynamic physiological system.

Rdcthera can offer two types of tumor mouse models for your customized biodistribution analysis, including syngeneic tumor mouse models and human xenograft mouse modes. Additionally, Rdcthera can perform biodistribution analysis in the mouse model provided by customers.

    Syngeneic Tumor Mouse Model

A most widely utilized preclinical model to evaluate anticancer therapeutics and agents.

    Human Xenograft Mouse Model

A tumor mouse model using human cell lines injected into immunocompromised hosts to evaluate the anti-tumor efficacy of your product.

  • Biodistribution Analysis Service
  • As a professional and experienced CRO company, Rdcthera can provide qualitative and quantitative biodistribution analysis services to promote your RDC project. The biodistribution analysis can be performed directly by injected to the tumor-bearing mice.

  • Imaging for Biodistribution Analysis
The biodistribution parameter is calculated by the PET imaging and SPECT imaging results.
  • Organ Collection for Biodistribution Analysis
The injected tumor-bearing mice is euthanized after specific times for radioactivity measurement and percentage injected dose (%ID) in each of the blood and tissue samples.
  • Autoradiography for Biodistribution Analysis
Autoradiographic analysis was performed for Vd (Apparent Volume of Distribution) determination by exposing the sections, fixed with acetone and sectioned on a cryostat, to a storage phosphor plate.
